We recently developed a robust, 7-day biofilm microplate protocol for creating biofilms with strongly adherent strains of Listeria monocytogenes, Escherichia coli O157:H7, and Salmonella serovars that could be used to examine the effectiveness of various commercial sanitizers. Listeria monocytogenes 99-38, E.coli O157:H7 F4546, and Salmonella Montevideo FSIS 051 were determined from prior studies to be good biofilm formers and could be recovered and enumerated from biofilms following treatment with trypsin.

Extended biofilms were generated by cycles of growth and washing daily, for 7 days, to remove planktonic cells. We examined five different sanitizers (three used at two different concentrations) for efficacy against the three pathogenic biofilms. ►▼ Show Figures Figure 1. Brashears, Ph.D. Shiga toxin-producing Escherichia coli (STEC) are pathogens of concern across various products within the food industry, as they have been connected to a wide variety of outbreaks and recalls. Most of the scientific literature concerning the removal of attached STEC cells focuses on E. coli O157:H7, as it was the first STEC to be considered an adulterant in nonintact beef products in the United States after a large outbreak from undercooked ground beef patties in 1982.[1] Worldwide, non-O157 STEC strains are estimated to cause 20 to 50 percent of STEC-related infections.[2] A review of outbreaks from 1983 through 2002 found six serogroups (O26, O111, O103, O121, O145 and O45) to be the most common non-O157 STECs causing human illness in the United States.[3] With an estimated 70 percent of non-O157 STEC infections being caused by these serogroups, the U.S.

Shown in Fig. 1 are populations of planktonic cells of E. coli O157:H7 strains 43895-EPS, ATCC 43895-, and ATCC 43895+ grown on HLJA at 22 and 12°C and treated with chlorine (10, 25, and 50 μg/ml) for 1, 3, 5, and 10 min. Cell suspensions contained various amounts of ECC (Table 1), depending on strain and growth conditions. Strain 43895-EPS was more resistant than strains ATCC 43895- and ATCC 43895+ to chlorine, indicating that protection was afforded by ECC.

The population of strain 43895-EPS grown at 22°C decreased by 5.2 log10 CFU/ml when treated for 10 min with 10 μg of chlorine/ml, but cells grown at 12°C were unaffected by the same treatment. Un nouvel exemple d’exclusion compétitive … Résumé. Nous avons étudié la possibilité d'utiliser un biofilm formé par des micro-organismes d'exclusion compétitive pour inactiver Escherichia coli O157:H7 sur une surface en acier inoxydable. Cinq micro-organismes présentant des activités inhibitrices vis-à-vis de E. coli O157:H7 ont été isolés à partir de graines de légumes et graines germées. Le micro-organisme avec la plus grande activité antimicrobienne a été identifié comme étant Paenibacillus polymyxa (souche T5).
