FOOD ANALYTICAL METHODS 06/03/21 Dilution of QuEChERS Extracts Without Cleanup Improves Results in the UHPLC-MS/MS Multiresidue Analysis of Pesticides in Tomato. Determination of Pesticide Residues by UHPLC-MS/MS The determination of the target pesticides in tomatoes using QuEChERS method without cleanup step was possible due to the good selectivity and sensitivity of the UHPLC-MS/MS system.
The mobile phase and gradient program promoted a proper separation of compounds. These conditions were already used in our laboratory by Martins et al. (2017) and de Matos et al. (2019), providing adequate analytical signals for all the compounds selected in our study. The chromatographic conditions provided high resolution and good peak shape for all the evaluated compounds.
UHPLC-MS/MS retention times (tR), SRM transitions, capillary voltages (CV), and collision energies (CE) were optimized for each analyte and are showed in Table 1. Sample Preparation Procedure In this work, sample preparation strategies based on QuEChERS method (original, acetate, and citrate versions) were evaluated based on recoveries and precision results. The reduction efficiency of the extracts of Albizia amara and Acacia concinna against most widely used organophosphorus pesticides such as dimethoate, malathion, dichlorvos, and chlorpyrifos on tomato samples was investigated.
The pesticide residues were quantified by gas chromatography with triple quadrupole mass spectrometer (GC‐MS/MS). The maximum reduction efficiency was achieved from the samples treated with 8% A. concinna extract on dichlorvos (87%) and dimethoate (84%) followed by 6% A. amara on malathion (83%) and chlorpyrifos (64%) and comparatively higher than water wash and it shows the reduction of only 14–38% for 15 min. The sensorial property and lycopene content of tomato samples were assessed and these plant extracts showed no significant effects on color, texture, and lycopene content after washing treatment. During the 2013 Florida Ag Expo, Dr. Gary Vallad, a plant pathologist with UF/IFAS, outlined some disease challenges tomato growers are facing in the absence of methyl bromide. Showing attendees a picture of a tomato field infested with classic fusarium wilt symptoms, he said this is becoming a more frequent sight across the state. “Back when I started with UF/IFAS in 2007, the only time I would see these yellow plants (fusarium wilt symptoms) was typically at the end of rows where too little methyl bromide went out because they turned off the fumigant too soon at the end of a row,” he said. “But starting three or four years ago, I began getting calls about these more uniformly diseased fields.” Another problem on the rise is fusarium crown/root rot.

Abstract A multiclass and multiresidue method for pesticide analysis in tomato was validated. Extraction and pre-concentration of the pesticide residues from acetonitrile extracts was performed by using dispersive liquid–liquid microextraction (DLLME) technique, followed by gas chromatography–mass detection.
