From the initial identification in 1942 in West Africa, PPR virus (PPRV) has spread throughout much of the developing world. PPRV is now considered endemic throughout Africa, with the notable exception of South Africa, the Middle-East and Israel, as well as South-, East-, and Central Asia. Despite this widespread dispersal, the evolution and transmission of PPRV in endemic populations is not well understood. This understanding will be critical in the planning of rational measures to eradicate PPRV by the planned time as defined by the FAO and OIE. PPR, also known as goat plague, is a highly contagious disease caused by PPR virus (PPRV) that infects small ruminants such as sheep and goats and can have a mortality rate as high as 90%. Cattle and pigs can be affected too, but tend not to develop clinical signs. First reported in 1942 in Western Africa’s Ivory Coast region, PPRV is now prevalent throughout Asia, the Middle East and Africa. It has been targeted for eradication by 2030 by the FAO (Food and Agriculture Organisation of the United Nations) and the OIE (World Organisation for Animal Health). This follows the successful eradication of the related disease rinderpest - a success that Pirbright scientists played a major role in achieving.

Despite control efforts in a number of countries, it has continued to spread across Africa and Asia, placing an increasing burden on the livelihoods of livestock keepers and on veterinary resources in affected countries. Given the similarities between PPR and rinderpest, and the lessons learned from the successful global eradication of rinderpest, the eradication of PPR seems appealing, both eliminating an important disease and improving the livelihoods of the poor in developing countries. We selected PPR disease to start with on the basis of two reasons viz. i) economic significance, ii) close relevance to its prototype, Riderpest (cattle plague) that has been effectively eradicated from the face of the earth. Current diagnostic tests for PPR involve the use of immunoassays to detect the antibodies against PPRV, virus isolation using adaptable cell lines and PPRV genome detection using RT-PCR or Real Time RT-PCR [13, 14, 15, 16, 17, 18]. However, these techniques require costly equipment and highly trained manpower that render these tests unsuitable for on-site application. Safety and efficacy were evaluated on goats and sheep in comparison with monovalent PPR and SGP vaccines. Goats were challenged by PPR virulent strain and sheep by SP virulent strain. The result shows that the combined PPR/SGP vaccine confers a good protection against both PPR and SGP infection with no significant difference with monovalent vaccines. The combined vaccine was used in the field on sheep flocks and good sero-conversion was detected for both diseases as soon as 14 days post vaccination. Keywords. These are often reported in sheep and goats populations and cause huge economic losses to the animal owners in developing countries. Detection of these diseases is routinely done using ELISA or microbiological methods which are being reinforced or replaced by molecular based detection methods including multiplex assays, where detection of different pathogens is carried out in a single reaction. In the present study, a one-step multiplex RT-qPCR assay was developed for simultaneous detection of Capripoxvirus (CaPV), Peste de petits ruminants virus (PPRV), Pasteurella multocida (PM) and Mycoplasma capricolum ssp. capripneumonia (Mccp) in pathological samples collected from small ruminants with respiratory disease symptoms.

Sheep and goat pox, along with peste des petits ruminants, are endemic throughout most of Africa, Asia and the Middle East. Whereas Rift Valley fever is endemic in Africa, outbreaks in the Middle East have been reported over the past decade, including the Arabian Peninsula. Saudi Arabia is a major importer of livestock, and understanding the prevalence of these viral infections would be useful for disease control.
