Developing vaccine technology platforms to respond to pandemic threats or zoonotic diseases is a worldwide high priority. The risk of infectious diseases transmitted from wildlife and domestic animals to humans makes veterinary vaccination and animal health monitoring highly relevant for the deployment of public health global policies in the context of “one world, one health” principles. Sub-Saharan Africa is frequently impacted by outbreaks of poultry diseases such as avian influenza and Newcastle Disease (ND). Here, an adenovirus-vectored vaccine technology platform is proposed for rapid adaptation to ND or other avian viral threats in the region.

Ethiopian isolates of the Newcastle Disease virus (NDV) were subjected to sequence and phylogenetic analyses, enabling the construction of antigenically matched vaccine candidates expressing the fusion (F) and hemagglutinin-neuraminidase (HN) proteins. ►▼ Show Figures Figure 1.

Maladie de Newcastle au Nigeria. CIRAD - AOUT 2016 - Le commerce de volailles vivantes au Mali : quel impact sur la propagation de la grippe aviaire et de la maladie de Newcastle ? Au Mali, l’aviculture a été promue ces quinze dernières années comme un moyen de lutter contre la pauvreté et d’augmenter l’apport de protéines alimentaires à faible coût. Son développement est cependant entravé par la présence endémique de la maladie de Newcastle et par la menace de l’influenza aviaire hautement pathogène (IAHP), responsable de foyers épidémiques dans plusieurs pays frontaliers.

Ces maladies se propagent en particulier sur les marchés de volailles vivantes, nombreux dans le pays. Pour mieux cerner ce risque, les chercheurs du Cirad et leurs partenaires ont entrepris une vaste étude sur les marchés de la moitié sud du pays ― une surface équivalente à celle de la France, qui rassemble 98 % des effectifs de volaille. Entre 2009 et 2010, ils ont recensé et visité tous les marchés où étaient vendues quotidiennement des volailles vivantes, soit 96 marchés répartis dans cinq régions et le district de la capitale, Bamako.

Open Access Highlights We describe the first nationwide survey of the seroprevalence of avian influenza and Newcastle disease viruses in backyard poultry in Oman. The composition of the backyard poultry sector in Oman is described. Both viruses were present in all regions of Oman. The bird-level seroprevalence of both viruses was high (∼40%). The flock-level seroprevalence of both viruses was very high (∼80%). The seroprevalence of AI was highest in turkeys, then chickens and ducks, and lowest in geese and guinea fowl. Significant risk factors were region (both viruses), seroprevalence of AIV (for NDV) and altitude (for AIV).

Abstract In Madagascar, Newcastle disease (ND) has become enzootic after the first documented epizootics in 1946, with recurrent annual outbreaks causing mortality up to 40%.

Four ND viruses recently isolated in Madagascar were genotypically and pathotypically characterised. By phylogenetic inference based on the F and HN genes, and also full-genome sequence analyses, the NDV Malagasy isolates form a cluster distant enough to constitute a new genotype hereby proposed as genotype XI. This new genotype is presumably deriving from an ancestor close to genotype IV introduced in the island probably more than 50 years ago. Our data show also that all the previously described neutralising epitopes are conserved between Malagasy and vaccine strains. However, the potential implication in vaccination failures of specific amino acid substitutions predominantly found on surface-exposed epitopes of F and HN proteins is discussed.

L'objectif de la thèse "Modélisation et contrôle de la transmission du virus de la maladie de Newcastle dans les élevages aviaires familiaux de Madagascar" était la modélisation, la validation et l'analyse mathématique de modèles de transmission de transmission du virus de la MN (VMN) dans les systèmes avicoles villageois en général et à Madagascar en particulier. Nous proposons de nouveaux modèles basés sur les connaissances actuelles de l'histoire naturelle de la transmission du VMN. Ainsi, nous présentons deux modèles mathématiques à compartiments de la transmission du VMN dans une population de poules : un premier modèle avec transmission environnementale et un deuxième modèle où la vaccination contre la maladie est prise en compte.
