The LD RPA assay was designed to amplify 160 nt of the kDNA gene, which is present in a high copy number in LD (~10,000 DNA copies/cell) .
The LD RPA assay detected down to 100 DNA copies applying the LD DNA linearized plasmid (Fig. 2), and one LD genomic DNA (Fig. 3). Using 96 buffy coats and skin biopsies collected from VL, asymptomatic and PKDL cases as well as negative control, the LD RPA assays had the same clinical sensitivity and specificity as the real-time PCR  (Table 3). However, the RPA produced results six to nine times faster than the real-time PCR, and does not require the same level of equipment and training. The most challenging step in the RPA assay development is the design of a primer pair able to amplify a very low DNA copy number [13, 17, 18, 25, 26, 27, 28].

Abstract This study investigated the efficacy of two collars for the treatment and prevention of flea infestations.
Additionally the effect of these collars on the incidence of Leishmania infantum infection as compared with a group of vaccinated dogs was evaluated. A total of 224 young dogs from private animal shelters were enrolled in April/May into four groups: G1, 55 dogs treated with 10% imidacloprid + 4.5% flumethrin collar (Seresto, Bayer Animal Health); G2, 60 dogs treated with 4% deltamethrin collar (Scalibor protector band, MSD Animal Health); G3, 54 dogs vaccinated with CaniLeish (Virbac Animal Health); and G4, 55 dogs left non-treated as controls.
Dogs were followed up at days 120 (September), 210 (December), and 360 (April-May). At those time points, clinical assessments, ectoparasite counts and blood, bone marrow and skin samples, to detect the presence of L. infantum, were performed. For this study, the amastigotes present in these insects were detected by histochemical and PCR techniques. Positive gene amplification for Leishmania was found in two horseflies of the species Tabanus importunus collected in the environment, and amastigote forms of Leishmania spp., as well as erythrocytes and leukocytes, were histochemically detected in one of that insect. The other analyzed insects were not positive by PCR our by direct parasitological examination. Leishmania spp. are protozoan parasites that cause a spectrum of pathologies in humans and other vertebrates, ranging symptomatically from cutaneous ulceration to visceral dissemination. In order to survive within the host, Leishmania are able to evade and modulate the host immune responses through the actions of their virulence factors; however, few putative virulence factors have been characterised during in vivo infection with Leishmania. The Leishmania major genome has revealed the presence of three peptide inhibitors of S1A family serine peptidases (ISPs), which are orthologues of a bacterial protease inhibitor, ecotin. This particular parasite species has previously been found only in Thailand and parts of Europe. No Leishmania infections of any species had been previously reported in a horse native to the United States. The discovery raises awareness of how widespread the parasite is and suggests a need for watchfulness regarding potential transmission to humans, the researchers said.
